Karolinska Institutet Researchers Identify Neural Causes of Rheumatoid Arthritis Pain Using AAV Technology
Researchers from the Karolinska Institutet in Stockholm, Sweden, have conducted a groundbreaking study investigating the neural basis of pain in rheumatoid arthritis (RA). Published in a peer-reviewed journal, the study challenges the conventional view that RA pain is solely driven by inflammation. Utilizing Adeno Associated Virus (AAV) products supplied by AMSBIO, the team conditionally knocked out the Ifnar1 gene in sensory neurons and inhibited the IFN1/MNK–eIF4E signaling pathway in mouse models. This precise approach allowed for the functional interrogation of joint-innervating neurons, avoiding the limitations of broader dorsal root ganglion manipulation. The results demonstrated significant alleviation of joint pain and restoration of dexterity and paw function in the subjects. The findings conclude that RA joint pain is caused by a specific cytokine and signaling pathway, offering promising avenues for targeted therapeutic interventions. This research highlights the potential of advanced viral delivery systems in uncovering complex disease mechanisms and developing more effective pain relief strategies for arthritis patients.
